"Enhancement of electroporation by enzyme treatment"

Grant in Aid of Research

Final Report: "The research project, “Enhancement of electroporation by enzyme treatment” was worked on by undergraduate student Bianca Williams-Ford during the 2011-12 academic year. Among the activities that Bianca did were preparation of media and growth of bacteria for electroporation, design of experiments to optimize electroporation, and comparison of published methods of electroporation. Other work on this project examined the effect of ampicillin pretreatment of E. coli on the efficiency of electroporation. Various times of incubation and concentrations of ampicillin were examined. The previously reported seven-fold increase in transformation due to ampicillin treatment remains the upper limit of improvement by this method. Further studies in the future will examine the effect of other treatments on the efficiency of transformation by electroporation. It is anticipated that another student may continue this project next semester. I am grateful to the Research Council for helping make this work possible."
